Elsa Lunghini est la fille de Georges Lunghini (comédien, photographe, compositeur) et de Christiane Jobert (artiste peintre et sculptrice). Elle est la nièce de Marlène Jobert et la cousine germaine d'Eva Green, de Joséphine Jobert et de Joy Green.
Âgée de sept ans, Elsa Lunghini interprète en 1981 son premier rôle au cinéma dans le film Garde à vue de Claude Miller, aux côtés de Romy Schneider, Lino Ventura et Michel Serrault.
